CJC-1295 No DAC & Ipamorelin

CJC-1295/Ipamorelin is a two-peptide research blend: 5MG of CJC-1295 No DAC and 5MG of ipamorelin in one vial. CJC-1295 No DAC, also called modified GRF 1-29 (mod GRF 1-29), is a tetrasubstituted analog of the first 29 residues of human growth hormone releasing hormone (GHRH), without the albumin-binding lysine, or drug affinity complex (DAC), of the named compound CJC-1295 (Jetté et al., 2005).

What is CJC-1295 No DAC & Ipamorelin?

CJC-1295/Ipamorelin is a two-peptide research blend: 5MG of CJC-1295 No DAC and 5MG of ipamorelin in one vial. CJC-1295 No DAC, also called modified GRF 1-29 (mod GRF 1-29), is a tetrasubstituted analog of the first 29 residues of human growth hormone releasing hormone (GHRH), without the albumin-binding lysine, or drug affinity complex (DAC), of the named compound CJC-1295 (Jetté et al., 2005). Ipamorelin is a synthetic pentapeptide that releases growth hormone through the receptor whose natural ligand is ghrelin, not the GHRH receptor (Raun et al., 1998; Kojima et al., 1999).

What the research covers

CJC-1295 No DAC is a modified GHRH(1–29) analog, while ipamorelin is a ghrelin-receptor secretagogue. The existing guide reports component studies in pituitary cells, animals and people, with no study combining these two peptides. The multi-day human half-life reported for CJC-1295 belongs to the albumin-binding DAC version, not No DAC. Separate receptor pathways provide a rationale for research on combinations; they do not establish an outcome or pharmacokinetic profile for this blend.

What is the CJC-1295 and ipamorelin blend?

The pair is studied because it acts through two separate receptor pathways, but every published study tested the components separately.

The PubChem entry for CAS 863288-34-0 also lists synonyms of the DAC version, so identify material by sequence and certificate. By moles the vial is not half and half: 5MG of CJC-1295 No DAC is about 1.5 micromoles and 5MG of ipamorelin about 7.0.

What the research covers

The literature falls into five areas. Each summary names its model; no study combined the two peptides.

CJC-1295 No DAC: a stabilized GHRH(1-29) analog

Native GHRH is cut at its amino terminus by a plasma dipeptidylaminopeptidase into a fragment with under one thousandth of its activity; its half-life in volunteers was 6.8 minutes (Frohman et al., 1986). The four substitutions target known weak points: the amino terminus against that cleavage, residue 8 against asparagine isomerization, residue 27 against methionine oxidation, and alanine at position 15 for receptor affinity.

Analogs designed this way were about three times as potent as native GHRH in rat pituitary cells and 11 to 13 times in pigs (Campbell et al., 1994). In ten healthy men, D-alanine at position 2 roughly halved the clearance of GHRH(1-29)-NH2 and lengthened its disappearance half-time from 4.3 to 6.7 minutes (Soule et al., 1994). The 5.8 to 8.1 day half-life reported in healthy adults belongs to the albumin-binding version only (Teichman et al., 2006). Models: human plasma, rat pituitary cells, pigs and healthy volunteers.

Ipamorelin: a selective ghrelin receptor agonist

Ipamorelin released growth hormone from primary rat pituitary cells with potency similar to GHRP-6, through a GHRP-like receptor, and in swine did not raise ACTH or cortisol beyond the levels seen with GHRH, even at over 200 times the amount giving a half-maximal growth hormone response, while GHRP-6 and GHRP-2 did (Raun et al., 1998).

In healthy men given 15-minute infusions at five levels, its terminal half-life was about 2 hours and growth hormone peaked once, near 40 minutes (Gobburu et al., 1999). A phase 2 trial in 114 adults after bowel resection found no significant difference from placebo in time to a tolerated solid meal (Beck et al., 2014). Models: rat pituitary cells, swine, healthy men and a phase 2 trial.

Two receptor pathways: why the classes are paired

In rat pituitary cells, GHRP-6 and GHRH at maximal concentrations released growth hormone synergistically. Only GHRH raised cyclic AMP, a GHRH antagonist did not block GHRP-6, and the authors concluded that the two act through different receptors (Cheng et al., 1989).

In 18 healthy men, submaximal amounts of GHRP-6 given with GHRH released growth hormone synergistically (Bowers et al., 1990); in a later study, small amounts of ghrelin given with GHRH did the same (Hataya et al., 2001). None used ipamorelin or CJC-1295 No DAC; the case for the blend is inferred from other members of each class. Models: rat cell culture and healthy men.

What has not been tested: the combination

No study indexed in PubMed has given CJC-1295 No DAC and ipamorelin together, in any model; an October 2026 search returned reviews and anti-doping methods only. A 2026 review says the pairing rests largely on anecdotal rationale; it puts the No DAC component in its lowest evidence tier, with no peer-reviewed human studies, and ipamorelin in its second tier, on phase 1 and postoperative ileus studies (Dominikowski et al., 2026). Nothing is published on the stability of the mixed powder, so handle it as a defined mixture with single-component controls.

Ingredient evidence ≠ blend evidence

The studies linked here may concern individual ingredients or related molecules. They do not establish that this named formulation has been tested as a whole.

What remains uncertain

A 2026 review says the pairing rests largely on anecdotal rationale; it puts the No DAC component in its lowest evidence tier, with no peer-reviewed human studies, and ipamorelin in its second tier, on phase 1 and postoperative ileus studies (Dominikowski et al., 2026). Each summary names its model; no study combined the two peptides.

The evidence label describes the linked research, not the suitability of a catalog product for use in people. Formulation, identity, purity, and measured content must be assessed separately.
